10 Aug Redefining the Treatment of Lymphatic Disease: Celltaxis Seeks to Transform One of Medicine’s Last Major Underserved Therapeutic Areas
Lymphatic diseases affect an estimated 250 million people worldwide, yet patients with lymphedema and lipedema still have no FDA-approved pharmaceutical treatment options. Celltaxis is seeking to change that by developing oral small-molecule therapies designed to restore lymphatic repair by addressing the underlying biology of disease.
Bench2Bedside recently sat down with Eileen Heffernan, Celltaxis’ Chief Executive Officer and co-founder, to discuss the Company’s mission to transform the treatment of lymphatic disease, the science behind its therapeutic approach, and why she believes Celltaxis is uniquely positioned to lead innovation in one of medicine’s most overlooked therapeutic areas.

Many people are unfamiliar with lymphatic disease. Why should they be paying attention now?
Eileen Heffernan:
Lymphedema and lipedema impact millions of people around the world, yet they remain among the most underdiagnosed and underserved chronic diseases in medicine. Patients have lived for decades with little more than compression garments, manual lymphatic drainage, and physical therapy because there simply haven’t been medicines designed to address the underlying disease.
We believe that’s beginning to change. Scientific understanding of lymphatic biology has advanced significantly, and we’re seeing growing recognition that chronic inflammation plays a central role in disease progression. That creates an opportunity to move beyond symptom management and begin treating the biology itself. For investors, it’s an opportunity to participate in what we believe could become an entirely new therapeutic category.

You’ve helped build Celltaxis since its founding, and your recent appointment as Chief Executive Officer reflects the Company’s next stage of growth. How do you view this transition?
Eileen Heffernan:
It’s an exciting evolution for the Company. I’ve been involved in shaping our strategy from the beginning, so becoming CEO isn’t about changing our mission. It’s about accelerating it.
Our goal has always been much bigger than advancing a single therapy. We want to establish Celltaxis as the company leading innovation in lymphatic disease. We have encouraging clinical data, a differentiated scientific approach, and a passionate team that believes patients deserve far better than the current standard of care.
The opportunity to help build a company that can truly change an overlooked field of medicine is incredibly motivating.

Celltaxis describes its therapeutic approach as restoring lymphatic repair rather than simply managing symptoms. What does that mean, and why is it such an important distinction?
Eileen Heffernan:
Today, patients with lymphedema are treated almost exclusively with treatments designed to manage the consequences of lymphatic dysfunction. This includes compression garments, manual lymphatic drainage and physical therapy. While those approaches can help reduce swelling and improve some quality of life, they don’t address the biological processes driving disease progression.
Celltaxis is pursuing a fundamentally different approach. Our oral small molecule therapies are designed to restore lymphatic repair by addressing one of the key, specific inflammatory pathways underlying disease progression. Scientific research has shown that excessive levels of leukotriene B4 (LTB4), a potent inflammatory signaling molecule, suppress the body’s natural ability to repair damaged lymphatic vessels. As those repair mechanisms become impaired, patients enter a cycle of chronic lymphatic dysfunction, contributing to swelling, fat deposition, tissue remodeling, fibrosis and declining patient function.
Our therapeutic approach selectively inhibits leukotriene A4 hydrolase (LTA4H), the enzyme responsible for producing LTB4. By reducing excessive LTB4 signaling, we’re seeking to restore the biological pathways responsible for lymphatic repair and regeneration, addressing an underlying driver of disease rather than simply managing its symptoms.

Your Phase 2 study produced strong data, but you’re now advancing a next-generation prodrug. Why?
Eileen Heffernan:
Our exploratory Phase 2 study generated improvements across multiple clinically meaningful measures, including limb volume, skin architecture, and patient quality of life. For us, those findings provide important clinical support for the underlying disease biology and the mechanism of action of our selective LTA4H inhibitor, reinforcing our belief that restoring lymphatic repair has the potential to redefine how these diseases are treated.
The next step was to ask how we could optimize that opportunity. Our prodrug is designed to improve pharmacokinetics of our oral small molecule therapy for sensitive cancer patients while preserving the same biological mechanism. It also strengthens our intellectual property and creates additional strategic and partnering opportunities as we build a broader franchise.

You’ve discussed potential synergies with GLP-1 therapies. How do you view that opportunity?
Eileen Heffernan:
Obesity and lymphatic dysfunction are closely connected, but weight loss alone doesn’t restore lymphatic function once disease is established. Our therapeutic approach addresses a different aspect of biology by promoting lymphatic repair.
Preclinical work suggests our approach may complement GLP-1 therapies, making this an exciting area for future development as more patients receive these medicines.

Where do you hope Celltaxis will be five to ten years from now?
Eileen Heffernan:
Of course, we want to bring transformative therapies to patients. But our ambition goes well beyond one product.
If we’re successful, we don’t want Celltaxis to be remembered simply as the company that developed an important therapy. We want to be known as the company that brought lymphatic disease out of the shadows and established it as a therapeutic area worthy of the same scientific attention as oncology, immunology or metabolic disease.
That’s the opportunity in front of us. It’s what energizes our team every day, and it’s why I’m so optimistic about what the future holds.
